This is an open-label, randomised controlled trial. The primary outcome was the change from baseline in ED medication (Viagra® and Cialis®) and total medication doses (Viagra® and Cialis®) on randomisation (Rx). The secondary outcomes were the change from baseline in ED medication (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) on Rx. The primary outcomes were changes from baseline in ED medication (Viagra® and Cialis®) and total medication doses (Viagra® and Cialis®) on randomisation (Rx).
A total of 999 patients were randomised (in 2, 3 or 4 groups) to either sildenafil (Viagra® and Cialis®) or placebo (placebo). The randomisation groups were double-blinded for one month. At the end of treatment, patients were assessed at 1, 4, and 12 months. The primary outcome was the change from baseline in total medication doses (Viagra® and Cialis®) and total medication doses (Viagra® and Cialis®) on randomisation (Rx). The secondary outcomes were the change from baseline in total medication doses (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) on Rx. There were 10 Rx in the placebo group and 5 Rx in the sildenafil group (at randomisation).
Patients were given a 12-lead telephone questionnaire, which was completed by all patients and assessed at the baseline ( baseline, at baseline, and at 1 and 4 months) and at the 2, 3, and 4 months. The patients completed the questionnaire at each month. All patients were asked to complete the following questionnaires:
Question 1:“Have you ever had erectile dysfunction in the last 12 months?”
Question 2:
Question 3:“Are you currently having any sexual activity and is it a routine activity?”
Question 4:“Are you currently using any form of phosphodiesterase type 5 (PDE5) inhibitor or currently using any form of phosphodiesterase type 5 (PDE5) inhibitor and are you on any other medications?”
The questionnaires were repeated at each month. The final questionnaires were completed in two-steps (1 and 2 steps) by all patients (in 2 steps).
Randomised 1-arm (in 2 steps) double-blinded for one month and randomised for 2 months. The trial protocol was registered with the Clinical Trial Registry for the Protection against Medicines. In each arm, two patients were given placebo (sildenafil) or Viagra® (Cialis®) and were also given an alternative treatment to Cialis®. The primary outcome was the change from baseline in ED medication (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) on Rx. The secondary outcomes were the change from baseline in ED medication (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) and the change from baseline in total medication doses (Viagra® and Cialis®) on Rx. Patients were given a single dose of each medication to which they were taking sildenafil and a single dose of placebo.
Randomised to sildenafil or placebo, patients in randomised to one of two treatment arms, either sildenafil or placebo.
1.Nitrates (e.g., nitroglycerin, isosorbide dinitrate): Cialis and nitrates both lower blood pressure. Taking them together can cause a significant and potentially life-threatening drop in blood pressure (hypotension), leading to dizziness, fainting, or even heart attack or stroke. It is contraindicated to use Cialis with nitrates or recreational "poppers" (amyl nitrites).
2.Alpha-blockers (e.g., prazosin, terazosin, tamsulosin): Both Cialis and alpha-blockers relax blood vessels, which can lead to excessive blood pressure reduction when used together. This may result in dizziness, fainting, or orthostatic hypotension (a sudden drop in blood pressure when standing). If needed, your doctor may adjust the dosage or recommend spacing the medications.
3.Other antihypertensive drugs (e.g., atenolol, propranolol, enalapril, lisinopril): Cialis can enhance the blood-pressure-lowering effects of these medications. While this interaction is usually less severe than with nitrates or alpha-blockers, it can still cause dizziness or fainting in sensitive individuals. Close monitoring of blood pressure is recommended.
Cialis (tadalafil) can interact with certain foods and beverages, potentially altering its effectiveness or increasing the risk of side effects. Here are the key drug-food interactions to be aware of:
